The second Madden game on the Game Boy Color features 31 NFL teams from the 2000 season, an All-Madden team, over 150 offensive and defensive plays, and three modes of play: Exhibition, Season, and Playoffs. This version also features new in-game colors as well as two-player link capabilities. After teams are selected, the action is played from an overhead view of a vertical field.

Two audibles can be assigned before the coin toss, allowing users to change their play at the line of scrimmage. Substitutions are also permitted during the game, and a no-huddle offense can be called to keep defenders on their toes. Quarter lengths can be set to 5, 10, or 15 minutes, with the default being 10.
